Product Specific Information

The monoclonal antibody 2D3 reacts with CD47, also known as integrin-associated protein (IAP) and neurophilin.
CD47 is a glycosylated five-transmembrane protein involved in cell adhesion through interactions with SIRP and integrins CD51/CD61 and CD41/CD61. It mediates bi-directional signaling affecting neural synaptic activity and macrophage phagocytosis. CD47 acts as a receptor for thrombospondin and is expressed in most hematopoietic and non-hematopoietic cells.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for use with normal human peripheral blood cells at 5 µL (0.6 µg)/test (final volume 100 µL; 10⁵–10⁸ cells/test)

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample to antibody cocktails prior to combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered to minimize spillover and spread impacts.

Target Information

CD47 (Integrin-associated protein, IAP) is a ubiquitous cell surface glycoprotein interacting with integrins and regulating signaling pathways. It modulates G-protein activation, intracellular calcium levels, and serves as a receptor for thrombospondin. CD47 prevents phagocytosis via interaction with CD172a (SIRPα) on macrophages and plays roles in cell migration, adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development.
