Product Specific Information

The monoclonal antibody 2D3 reacts with CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein involved in adhesion via interactions with SIRP and integrins (CD51/CD61, CD41/CD61). It mediates bi-directional signaling affecting neural synaptic activity and macrophage phagocytic regulation. CD47 serves as a receptor for thrombospondin and modulates T cell activation or apoptosis depending on thrombospondin levels.

Expression is found in most hematopoietic cells (T, B cells, monocytes, platelets, erythrocytes) and non-hematopoietic cells.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use at 5 µL (0.6 µg) per test (100 µL final volume). Recommended cell range: 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample to antibody cocktails before combining with cells.
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor Conjugates: Based on Phiton™ technology using nucleic acid dye structures engineered for optimized fluorescent performance and minimized spillover.

Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitous transmembrane glycoprotein that interacts with integrins, modulating signaling pathways such as G protein activation and calcium influx during cell adhesion. It is a receptor for thrombospondin and plays roles in membrane transport and signal transduction. CD47 expression is broad but reduced on Rh erythrocytes. Interaction with CD172a (SIRPα) inhibits macrophage phagocytosis of CD47-positive cells. The CD47–CD172a system affects cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, especially in synapse-rich brain and retina regions.
