Product Specific Information

Description:
The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a member of the TNFR family and is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype-switching and, in combination with IL-4, protects B cells from apoptosis and promotes proliferation. Interaction of CD40 with CD154 (gp39) on T cells is critical for T-B cell crosstalk and immune regulation.
5C3 is reported to activate B cells in in vitro functional assays.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤1 µg per test (100 µL final volume). Cell number may vary from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information

CD40 antigen is a single-chain glycoprotein of the TNF/NGF superfamily, homologous to CD30. Expressed on all B cells except plasma cells, and also on epithelial cells, carcinomas, and lymphoid dendritic cells.
CD40 is essential for immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
AKNA transcription factor regulates CD40 expression; TNFR2 acts as a signal transduction mediator.
CD40-CD154 interaction is implicated in amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis.
