Description:
The HM40-3 monoclonal antibody reacts with mouse and rat CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a member of the TNFR family and is expressed by mouse B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. CD40 regulates B cell development and maturation by inducing Ig isotype switching, protecting B cells from apoptosis, and promoting proliferation. Interaction with CD154 (gp39) is critical for T-B cell communication and immune regulation.
Applications Reported:
The HM40-3 antibody is reported for use in flow cytometry and immunohistochemistry. It can inhibit soluble CD154 binding to CD40 and activate B cells in vitro. For functional assays, use Functional Grade purified HM40-3 (Product # 16-0402).
Applications Tested:
Tested by flow cytometry using mouse splenocytes. Recommended ≤1 µg/test in 100 µL final volume. Optimize concentration empirically for best results.

Target Information
CD40 is a single-chain glycoprotein, a member of the TNF/NGF receptor superfamily, homologous to CD30. It is expressed on all B cells except plasma cells, as well as on some epithelial cells, carcinomas, and dendritic cells. CD40 is essential for immune and inflammatory responses, including T cell–dependent immunoglobulin class switching, memory B cell development, and germinal center formation. It interacts with adaptor protein TNFR2 and is involved in amyloid-beta-induced microglial activation, potentially linked to Alzheimer’s disease. Mutations in CD40 are associated with Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
