The 1C10 monoclonal antibody reacts with mouse CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 belongs to the TNFR family and is expressed by mouse B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells.

CD40 regulates B cell development and maturation by inducing Ig isotype switching. Together with IL-4, it protects B cells from surface Ig-induced apoptosis and promotes proliferation. Interaction of CD40 with its ligand CD154 (gp39) on T cells is important for T-B cell crosstalk and plays a role in costimulation and immune regulation.

The monoclonal antibody 1C10 is reported to have agonistic activity both in vitro and in vivo.

Applications Reported: Used in flow cytometric analysis, immunoprecipitation, and immunohistochemical staining of frozen tissue sections. For functional assays, use Functional Grade purified 1C10 (Product #16-0401-82).

Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤1 µg per test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.

Target Information

CD40 is a single-chain glycoprotein of the TNF/NGF receptor superfamily, homologous to CD30. It is expressed on all B cells except plasma cells, and also on certain epithelial cells, carcinomas, and lymphoid dendritic cells.

CD40 is essential for immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. Transcription factor AKNA regulates CD40 expression, facilitating cell interactions. TNFR2 acts as an adaptor in CD40 signaling.

CD40-CD154 interaction is implicated in amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis. Two alternatively spliced transcript variants of CD40 encoding distinct isoforms have been reported.
