Product Specific Information
The HIP8 monoclonal antibody reacts with the human CD41 molecule (integrin alphaIIb, platelet GPIIb). CD41 non-covalently associates with integrin beta3 (CD61) and is expressed by megakaryocytes and platelets. The CD41/CD61 complex functions as a receptor for fibronectin, fibrinogen, von Willebrand factor, vitronectin, and thrombospondin, mediating platelet aggregation. HIP8 blocks platelet aggregation.
Applications Reported: Flow cytometric analysis and immunohistochemical staining.
Applications Tested: Flow cytometric analysis of human platelets at ≤0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Antibody titration recommended for optimal assay performance.
Purity: >90% (SDS-PAGE).
Aggregation: <10% (HPLC).
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD41 (platelet glycoprotein IIb, ITGA2B) is composed of two subunits—120 kDa alpha and 23 kDa beta—that interact with CD61 in the presence of calcium to form a functional adhesive receptor. It mediates platelet aggregation and binds von Willebrand factor, fibrinogen, fibronectin, and vitronectin upon vascular injury. CD41 is expressed on megakaryocyte-platelet lineage and during early hematopoietic differentiation.
Diseases associated with CD41 dysfunction include Glanzmann Thrombasthenia and Platelet type-16 bleeding disorder.
