Product Specific Information

The ICRF44 monoclonal antibody reacts with human CD11b, a 165 kDa adhesion molecule associated with integrin beta2 (CD18). It is expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. CD11b functions as a receptor for ICAM family members (CD54, CD102, CD50) and iC3b, playing a crucial role in cell-cell and cell-matrix interactions.

Applications Reported: Flow cytometric analysis.

Applications Tested: Pre-diluted and tested by flow cytometry using normal human peripheral blood cells. Recommended use is 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Use a 660/20 bandpass filter. Protect from light due to photo-induced oxidation sensitivity.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333).

Target Information

CD11b (integrin alpha-M, ITGAM) forms a heterodimer with CD18 (integrin beta-2) known as Mac-1 or CR3. It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia. CD11b/CD18 binds ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating cell adhesion and immune response. Expression varies with cell type and maturation state and is a key microglial marker.
