Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. Complexes of LPS and LBP bind with high affinity to monocytes through surface CD14.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry on normal human peripheral blood cells. Recommended use: 5 µL (0.5 µg) per test in 100 µL final volume.

Fluorochrome Information:
Super Bright 645 is a tandem dye excited with the violet laser (405 nm) and emits at 645 nm. Recommended detection filter: 660/20 bandpass.

Staining Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light due to photo-induced oxidation sensitivity.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness and compensation.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on mature monocytes, macrophages, and neutrophils. It functions as a multifunctional lipopolysaccharide receptor and exists in both membrane-bound and soluble forms. CD14 interacts with lipopolysaccharide-binding protein (LBP) and plays a role in immune response modulation, including recognition of apoptotic cells and mediation of inflammatory processes.
Diseases associated with CD14 dysfunction include Mycobacterium chelonae infection and Croup.
