Product Specific Information

The Sa2-8 monoclonal antibody reacts with mouse CD14, a 53–55 kDa GPI-linked glycoprotein that serves as a receptor for LPS-LBP complexes. It associates with TLR4 and participates in signaling and response to bacterial LPS.
CD14 is expressed on macrophages and can also be found in serum in secreted form. Sa2-8 has weak antagonistic activity in NF-κB activation or TNF-α production upon LPS stimulation.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Validated on thioglycolate-elicited peritoneal macrophages at ≤0.5 µg/test. One test corresponds to staining a sample of 10⁵–10⁸ cells in 100 µL final volume. Optimize antibody titration for best results.

Super Bright 645 is a tandem dye excited at 405 nm and emits at 645 nm. Recommended detection with a 660/20 bandpass filter. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, employ Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Fixation: Samples can be stored up to 3 days at 4°C in the dark using:

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It acts as a multifunctional receptor for lipopolysaccharides and is found in both membrane-bound and soluble forms. CD14 interacts with LBP and lipoprotein particles to neutralize bacterial LPS and can bind apoptotic cells, influencing allergic, inflammatory, and infectious responses.
Alternative splicing produces multiple transcript variants encoding the same CD14 isoform. Associated diseases include Mycobacterium chelonae infection and croup.
