Product Specific Information

The M5/114.15.2 monoclonal antibody recognizes mouse MHC class II molecules encoded by both I-A and I-E subregions (I-Aᵇ, I-Aᵈ, I-Aᵠ, I-Eᵈ, I-Eᵏ, but not I-Aᶠ, I-Aᵏ, or I-Aˢ). It detects polymorphic determinants on B cells, monocytes, macrophages, dendritic cells, and activated T lymphocytes from mice with H-2ᵇ, H-2ᵈ, H-2ᵠ, H-2ᵖ, H-2ʳ, and H-2ᵘ haplotypes, but not from H-2ˢ or H-2ᶠ mice.
This antibody can inhibit I-A–restricted T cell responses in certain haplotypes (H-2ᵇ, H-2ᵈ, H-2ᵠ, H-2ᵘ).

Applications Reported:
Flow cytometry, immunoprecipitation, Western blot, immunohistology (frozen and paraffin), and in vitro blocking assays.
For functional assays, use Functional Grade purified M5/114.15.2 (Product #16-4321-82).

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes at ≤0.125 µg/test (100 µL reaction volume). Recommended to titrate for optimal performance.

Target Information

MHC class II molecules are transmembrane glycoproteins expressed on professional antigen-presenting cells (macrophages, dendritic cells, B cells). They bind endocytosed antigens and present them to T cells via an open-ended antigen-binding groove (15–24 amino acids long).
HLA and MHC antibodies are key tools in immunopeptidomics, enabling identification and characterization of neoantigens using LC–MS/MS.
