Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein. Two isoforms (345 and 395 kDa) are expressed in dividing cells. Ki-67 is detected during active phases of the cell cycle (G1, S, G2, and mitosis) but absent in resting cells (G0).
During interphase, Ki-67 localizes to the nucleus and redistributes to chromosomes during mitosis, associating with heterochromatin-bound proteins such as CBX3.
In tumor studies, Ki-67 serves as a marker for determining the fraction of proliferating cells.

This antibody also recognizes canine Ki-67.

Applications Reported: Immunocytochemistry and immunohistochemistry on formalin-fixed paraffin-embedded tissues.
Applications Tested: ICC on paraformaldehyde-fixed MCF7, MDCK, and MDA-MB-175 cell lines; IHC on FFPE human skin using citrate buffer antigen retrieval.
Recommended concentration ≤0.5–1 µg/mL, with titration for optimal performance.

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It associates with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.
Ki-67 shows cell cycle-dependent localization: perinucleolar in G1, nuclear matrix in G2, and chromosomal in M phase.
It is widely used as a proliferation marker since it is absent in G0 cells but increases through mitosis.
Ki-67 antibodies are useful for determining the growing fraction in neoplasms and have prognostic value comparable to thymidine-labelling index.
Commonly used as a neuronal marker for cell cycling and proliferation.
