Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha).
CD127 forms a complex with CD132 (common gamma chain, γc) to create the functional IL-7 receptor (IL-7R).
It is a type I glycoprotein (75–80 kDa) expressed on immature B cells, thymocytes, and most mature T cells, with transient downregulation upon activation.

Functional Role:

• Mediates IL-7 signaling via Jak/STAT pathways
• Essential for lymphocyte development and T-cell homeostasis
• Regulates TCR locus rearrangement in γδ T cells

Regulatory T Cell Marker:
CD127 expression is downregulated on CD4⁺CD25⁺ regulatory T cells (Tregs).
Cells with low CD127 and high CD25 are enriched for Foxp3⁺ Tregs with suppressive activity.

Blocking and Controls:
Binding to PBMCs is blocked by pre-incubation with recombinant human IL-7 (Product #14-1079-80).

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested for use at 5 µL (0.4 µg)/test in 100 µL final volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Product #C001T06F01) for optimal staining.
  • Add 5 µL per sample (regardless of the number of NovaFluors).
  • For single-color controls, use 5 µL per 100 µL of cell sample.
