Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 forms a complex with CD132 (common gamma chain) to create the IL-7 receptor (IL-7R), a type I glycoprotein (75–80 kDa). It is expressed by immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

IL-7 binding triggers signal transduction via tyrosine kinase pathways, including Jak/STAT. IL-7 is essential for lymphocyte development and T-cell homeostatic proliferation. CD127 expression is down-regulated in CD4⁺CD25⁺ regulatory T cells (Tregs), making CD127⁻CD4⁺CD25⁺ a reliable marker for Tregs expressing Foxp3.

Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7 (Product #14-1079-80).

Applications Tested: Pre-titrated and validated for flow cytometry at 5 µL (0.6 µg)/test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability detection.
• Use CellBlox Plus Blocking Buffer (Product #C001T06F01) for optimal staining. Add 5 µL per sample before combining with cells.

Target Information

CD127 (Interleukin-7, IL-7) is a glycoprotein regulating lymphopoiesis. It acts via the IL7R alpha/gamma heterodimer. The gamma(c) chain activates signal transduction, while the alpha chain mediates specific signaling. CD127 promotes proliferation of precursor B cells, thymocytes, and mature T cells. Dysfunction is associated with severe combined immunodeficiency (SCID).
