Product Specific Information

The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor belonging to the immunoglobulin gene superfamily. The N-terminal amino acid sequence (aa1–107) shows high homology to Ig kappa light chains, while the carboxyl-terminal region is proline-rich, forming a stalk-like structure projecting the Ig domain.

CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, natural killer cells, a small subset of normal B cells, and malignant B cells. Cross-linking of surface CD7 enhances T cell and NK cell activity, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 directly associates with phosphoinositol 3′-kinase, and ligation induces D-3 phosphoinositide production and tyrosine phosphorylation.

A subpopulation of human CD34(+) CD38(-) cord blood cells expressing CD45RA, HLA-DR, and high CD7 levels has lymphoid progenitor capacity, generating B cells, NK cells, and dendritic cells but not myeloid or erythroid cells. CD7(+) phenotype distinguishes primitive lymphoid progenitors from pluripotent stem cells. CD7 may cooperate with CD28 in Treg function; deficiency in both reduces Treg numbers and activity.

Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.25 µg)/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample prior to combining with cells.
  • For single-color controls, use 5 µL CellBlox per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures optimized for spillover and spread management.
