Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 forms a complex with CD132 (common gamma chain) to create the IL-7 receptor (IL-7R), a type I glycoprotein (~75–80 kDa). It is expressed on immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

IL-7 binding triggers signaling through tyrosine kinase pathways, including Jak/STAT, essential for lymphocyte development and T-cell homeostasis. CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs), making CD127 a key marker for identifying true Treg populations with Foxp3 expression and suppressive function.

Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7 (Product #14-1079-80).

Each product includes:

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry of normal human peripheral blood cells. Recommended use: 5 µL (0.4 µg) per test in 100 µL final volume, with 10⁵–10⁸ cells/test.

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Product #C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor conjugates use Phiton™ technology with engineered fluorescent signatures designed to minimize spectral spillover and spread.

Target Information

CD127 (Interleukin-7 Receptor alpha, IL-7Rα) is a glycoprotein involved in lymphopoiesis regulation. The active IL-7 receptor is a heterodimer of alpha and gamma chains. The gamma(c) chain activates signal transduction, while the alpha chain mediates specific signaling. CD127 promotes proliferation of precursor B cells, thymocytes, and mature T cells.
Defects in CD127 function are associated with severe combined immunodeficiency (SCID).
