Description:
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 forms a complex with CD132 (common gamma chain) to create the IL-7 receptor (IL-7R), a type I glycoprotein (75–80 kDa). It is expressed on immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

IL-7 binding triggers signal transduction through tyrosine kinase pathways, including Jak/STAT. IL-7 is essential for lymphocyte development and T-cell homeostatic proliferation. CD127 signaling also regulates TCR locus rearrangement in γδ T cells.

CD127 expression is notably down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs). The CD4⁺CD25⁺CD127⁻ population is highly enriched for regulatory T cells expressing Foxp3 and exhibiting suppressive activity.

Binding of eBioRDR5 to PBMCs can be blocked by pre-incubation with recombinant human IL-7 (Product #14-1079-80).

Each product contains:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for normal human peripheral blood cells. Use 5 µL (0.2 µg)/test in 100 µL final volume with 10⁵–10⁸ cells/test.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Product #C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample prior to cell incubation. For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology utilizing engineered nucleic acid dye structures for optimized fluorescence signatures with reduced spillover and spread.

Target Information

CD127 (Interleukin-7 Receptor alpha, IL7Rα):
A glycoprotein involved in lymphopoiesis regulation. The active receptor is a heterodimer of alpha and gamma chains. The gamma chain activates signal transduction, while the alpha chain mediates specific signaling events. CD127 promotes proliferation of precursor B cells, thymocytes, T-cell progenitors, and mature CD4⁺/CD8⁺ T cells.
Defects in CD127 function are associated with severe combined immunodeficiency (SCID).
