Target Information
CD178 (Fas ligand, FasL)은 type-II 막 단백질로 세포질 내 N-말단과 세포외 공간으로 확장된 C-말단을 가집니다. 수용체 FasR은 TNF 및 NGF 수용체 계열에 속하는 type-I 막 단백질입니다.
CD178은 FasR과 결합 시 apoptosis를 유도하며, 막 결합형(45 kD)과 용해형(26 kD) 형태로 존재할 수 있습니다. 용해형 단백질은 metalloproteinase에 의해 절단되어 세포로부터 방출됩니다.
CD178–Fas 상호작용은 수용체의 올리고머화를 유도하고, 세포 사멸 신호를 전달합니다. CD178은 주로 활성화된 T-림프구와 NK 세포에서 발현되며, 고환 및 안구와 같은 면역 특권 부위에서도 발현됩니다. 또한 흉선, 간, 난소, 폐, 심장, 신장 등 다양한 조직에서도 보고되었습니다.
CD178을 통한 apoptosis 유도는 항바이러스 면역 반응에 중요한 역할을 하며, FAS/CD178 시스템은 AIDS, 간염, 암 등 여러 질병에 관여하는 것으로 알려져 있습니다. 또한 CD178 농도는 죽상동맥경화증 및 염증성 질환과 관련이 있는 것으로 보고되었습니다.
