Product Specific Information

This 4MEJJ monoclonal antibody recognizes mouse aryl hydrocarbon receptor (AHR), a ligand-activated transcription factor mediating toxic responses to environmental contaminants such as PCB and TCDD. AHR also binds naturally occurring compounds from fruits, vegetables, and metabolic processes. It is cytoplasmic in complex with HSP90, p23, and XAP2/AIP/ARA9, and translocates to the nucleus upon ligand binding to regulate gene expression (e.g., CYP1A1).

The antibody recognizes b-1, b-2, and d alleles of mouse AHR but not human AHR.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested in intracellular staining of TH17-polarized mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Recommended use ≤1 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

eFluor® 660 replaces Alexa Fluor® 647. Excitation: 633–647 nm; Emission: 668 nm; Laser: Red.

Target Information

AHR (Ah Receptor) is part of a family including ARNT (HIF-1 Beta) and Drosophila PER/SIM proteins. It contains an N-terminal PAS domain and binds specific DNA enhancer sequences to initiate transcription of cytochrome P450 (CYP1A1). AHR regulates xenobiotic-metabolizing enzymes and responds to aromatic hydrocarbons. In resting cells, AHR complexes with HSP90 in the cytoplasm; upon ligand binding, it translocates to the nucleus forming a heterodimer with ARNT for DNA binding.
Associated diseases include eosinophilic fasciitis and seborrheic dermatitis.
