Product Specific Information
This 5H12 monoclonal antibody reacts with mouse Autoimmune Regulator (AIRE), a 58-kDa transcription factor highly expressed in mature medullary thymic epithelial cells (mTECs). Although AIRE mRNA is detected in peripheral tissues such as lymph nodes and dendritic cells, protein expression outside the thymus remains unclear.
AIRE is involved in thymocyte development and regulation of tissue-specific antigen expression during negative selection. It also contributes to chemokine expression and antigen presentation.
The 5H12 antibody recognizes mouse AIRE isoforms 1 and 2.

Used for intracellular flow cytometric analysis and immunohistochemical staining of frozen tissue sections.
Applications Tested:
Tested by immunohistochemistry on frozen 5-week-old mouse thymus; recommended ≤10 µg/mL. Optimize dilution for specific assays.

Target Information
AIRE is a transcriptional regulator that binds DNA as a dimer or tetramer and regulates gene expression. It is a nuclear protein composed of 545 amino acids, containing an HSR domain, two PHD-type zinc fingers, and a SAND domain. It may form homodimers, homotetramers, and nuclear bodies, interacting with coactivators CBP and CREBBP.
Three isoforms of AIRE are reported, one potentially involved in nuclear-mediated decay (NMD). AIRE mediates E3 ubiquitin ligase activity and plays a critical role in immune regulation. Mutations in AIRE cause autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy (APECED).
