Product Specific Information

The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat FOXP3 (FORKHEAD BOX P3, SCURFIN, JM2). FOXP3 is a 49–55 kDa transcriptional regulator associated with regulatory T cells (Treg). This antibody recognizes amino acids 75–125 of mouse FOXP3.

The antibody is optimized for use with the Foxp3/Transcription Factor Buffer Staining Set (Product #00-5523-00). Other fixation buffers are not recommended.

Applications Reported:

• Intracellular staining and flow cytometric analysis
• Immunohistochemical staining of frozen tissue sections

Applications Tested:

• Intracellular staining and flow cytometry of mouse splenocytes using the Foxp3/Transcription Factor Buffer Set (≤0.5 µg/test)
• Immunohistochemistry of frozen mouse tissue (≤20 µg/mL)
• Recommended to titrate for optimal performance

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator crucial for immune homeostasis. It is highly expressed in CD4⁺CD25⁺ regulatory T cells and absent in CD8⁺ T cells. FOXP3 mutations cause immune dysregulation syndromes such as IPEX in humans and scurfy phenotype in mice. Multiple isoforms exist due to alternative splicing, and expression may be regulated by p53 in certain cancer cells.
