Product Specific Information
The eBioH4A3 monoclonal antibody reacts with human CD107a (LAMP-1), a highly glycosylated ~110 kDa protein expressed mainly on lysosomal/endosomal membranes. CD107a transiently appears on the surface of degranulating cytolytic T cells, activated platelets, and some cancer cells.
Applications Reported:
Suitable for intracellular staining and flow cytometric analysis.
Applications Tested:
Pre-titrated and validated for intracellular staining and flow cytometric analysis of Jurkat cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Recommended use: 5 µL (0.25 µg) per test (100 µL total volume, 10⁵–10⁸ cells/test).
Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emitting at 445 nm. Ensure instrument compatibility with the Violet laser.
Filtration:
0.2 µm post-manufacturing filtered.
Target Information
LAMP1 (CD107a) is a major lysosomal membrane component, with 1–2% also found on the plasma membrane. It is a heavily glycosylated type I membrane protein involved in lysosome biogenesis and fusion with phagosomes. LAMP1 relocation to the plasma membrane depends on a carboxyl-terminal tyrosine-based motif (YXXI). Mutations in this motif disrupt lysosomal localization.
Cell surface LAMP1 promotes adhesion of PBMCs to vascular endothelium, suggesting a role in inflammatory cell recruitment. Increased LAMP1 expression is observed in neurons and glial cells near senile plaques in Alzheimer's disease and in activated immune cells during thymic involution. LAMP1 serves as a reliable marker of mast cell activation.
