Product Specific Information

The eBioH4B4 monoclonal antibody reacts with human CD107b (LAMP-2), a highly glycosylated type I transmembrane protein (~105 kDa). It is expressed in lysosomal/endosomal membranes of most cells, on degranulating T cells, activated platelets, and certain cancer cells.
Mutations in CD107b cause Danon disease, a lysosomal glycogen storage disorder.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated using normal human peripheral blood cells with the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00). Recommended usage: 5 µL (0.25 µg) per test in 100 µL total volume. Cell counts typically range from 10⁵–10⁸ cells/test.

Fluorophore Information:
eFluor 450 is an alternative to Pacific Blue, emitting at 446 nm and excited at 405 nm (violet laser). Ensure instrument compatibility with this fluorochrome.

Target Information

LAMP2 (CD107b) is a lysosome-associated membrane glycoprotein involved in lysosomal biogenesis, fusion with phagosomes, and chaperone-mediated autophagy.
It plays roles in tumor cell metastasis, cellular adhesion, and protection.
Mature LAMP2 is extensively glycosylated (100–130 kDa).
Variants include LAMP-2A, -2B, and -2C.
Deficiency leads to accumulation of autophagosomes and Danon’s disease (cardiomyopathy, myopathy).
