Cyclin D1 (PRAD-1, bcl-1)는 세포주기 조절에 중요한 역할을 하는 단백질로, cdk4 및 cdk6과 결합하여 Rb 단백질을 인산화시킵니다. 이 단백질은 다양한 인간 종양에서 과발현되며, 세포 증식 및 분화 조절에 관여합니다.
Cyclin D1은 G1 단계에서 합성되어 성장 인자 자극에 따라 CDK4 또는 CDK6과 복합체를 형성합니다. Cyclin-CDK 복합체는 Rb 단백질의 성장 억제 기능을 불활성화하고 CDK 억제제(p21Cip1, p27Kip1)를 조절합니다. 성장 인자가 없을 경우 Cyclin D1은 불안정하며, 인산화에 의해 유비퀴틴 매개 분해를 겪습니다.
Cyclin D1은 세포주기 G1/S 전환에 필수적인 조절 단위로 작용하며, 종양 형성과 관련된 연구에 유용한 표적입니다.
