Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding to estrogen, undergoes a conformational change enabling dimerization and DNA binding to estrogen response elements (ERE). ER regulates gene transcription through coactivator or corepressor recruitment. Two forms exist—alpha (ESR1) and beta (ESR2)—with multiple alpha isoforms (66, 53, 47, and 35 kDa) due to alternative splicing. ERs are widely expressed and essential for sexual development, reproductive function, and bone health. They are implicated in breast and endometrial cancers and osteoporosis.
