Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a member of the TNFR family and is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype-switching and, together with IL-4, prevents apoptosis and promotes proliferation.

Interaction of CD40 with CD154 (gp39) on T cells is critical for T-B cell crosstalk and immune regulation. 5C3 is also used for activation of B cells in in vitro functional assays.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometry on normal human peripheral blood cells. Recommended ≤5 µL (0.6 µg)/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best results.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
    • Add 5 µL per sample or per 100 µL of cell sample for single-color controls.

NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures, providing engineered fluorescence with minimized spillover.

Target Information

CD40 antigen is a single-chain glycoprotein, member of the TNF/NGF superfamily, homologous to CD30. It is expressed on all B cells except plasma cells and also on epithelial cells, carcinomas, and dendritic cells. CD40 mediates immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.

AKNA transcription factor regulates CD40 expression, while TNFR2 adaptor protein mediates CD40 signaling. CD40-CD154 interaction is essential for amyloid-beta-induced microglial activation, linked to Alzheimer’s disease pathogenesis. Two alternatively spliced transcript variants exist. CD40 dysfunction is associated with Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
