Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a TNFR family member expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells.
It regulates B cell development and maturation, induces Ig isotype-switching, and protects B cells from apoptosis while promoting proliferation. Interaction with CD154 (gp39) is critical for T-B cell crosstalk and immune regulation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Tested: Tested on normal human peripheral blood cells at ≤5 µL (0.6 µg)/test.
A test corresponds to staining a cell sample (10⁵–10⁸ cells) in 100 µL final volume. Optimize antibody titration per assay.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• Always use CellBlox Plus Blocking Buffer for optimal staining. Add 5 µL per sample (regardless of NovaFluor number in panel).
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD40 is a single-chain glycoprotein, part of the TNF/NGF receptor superfamily, homologous to CD30. Expressed on all B cells except plasma cells, and also on some epithelial cells, carcinomas, and dendritic cells.
It mediates immune and inflammatory responses, including T cell-dependent class switching, memory B cell development, and germinal center formation.
CD40 interaction with CD40L is implicated in amyloid-beta-induced microglial activation, linked to Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Associated diseases include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
