Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein belonging to the TNFR family. CD40 is expressed on B lymphocytes, follicular dendritic cells, thymic epithelium, and some peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype-switching and, with IL-4, prevents apoptosis and promotes proliferation.

Interaction between CD40 and its ligand CD154 (gp39) is critical for T–B cell crosstalk and immune regulation. The 5C3 antibody can activate B cells in vitro functional assays.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Tested

Pre-titrated and tested for flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.5 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Compatibility Notes

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays with NovaFluor dyes.

CellBlox Plus Blocking Buffer

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01).
Add 5 µL per sample for optimal staining. Use in all experiments involving NovaFluor conjugates.

NovaFluor Dye Technology

NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures engineered for reduced spectral spillover and spread.
Excitation: 494 nm; Emission: 585 nm; Laser: 488 nm (Blue).

Target Information

CD40 is a single-chain glycoprotein of the TNFR/NGF superfamily, homologous to CD30. It is expressed on all B cells except plasma cells and also on epithelial cells, carcinomas, and lymphoid dendritic cells.

CD40 mediates diverse immune and inflammatory responses including:

  • T cell-dependent immunoglobulin class switching
  • Memory B cell development
  • Germinal center formation

Regulated by transcription factor AKNA and adaptor protein TNFR2. The CD40–CD40L interaction is implicated in amyloid-beta-induced microglial activation, associated with Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM Immunodeficiency and CD40 ligand deficiency.
