Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a member of the TNFR family and is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype-switching and protecting B cells from apoptosis in combination with IL-4.

Interaction of CD40 with CD154 (gp39), its ligand on T cells, is crucial for T-B cell crosstalk, costimulation, and immune regulation. 5C3 is reported to activate B cells in in vitro functional assays.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry on normal human peripheral blood cells
Recommended Use: 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample.

Technology: NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures to minimize spillover and spread.

Target Information

CD40 is a single-chain glycoprotein, member of the TNFR/NGFR superfamily, homologous to CD30. It is expressed on all B cells except plasma cells, and also on some epithelial cells, carcinomas, and dendritic cells. CD40 mediates immune and inflammatory responses including:

  • T cell-dependent immunoglobulin class switching
  • Memory B cell development
  • Germinal center formation

AKNA transcription factor regulates CD40 expression, and TNFR2 acts as a signal transduction mediator. CD40-CD154 interaction is implicated in amyloid-beta-induced microglial activation, potentially related to Alzheimer’s disease pathogenesis.

Diseases associated with CD40 dysfunction include:

  • Type 3 Hyper-IgM immunodeficiency
  • CD40 ligand deficiency
