Purity
No colonies form after inoculation of LB-agar medium containing 100 µg/ml ampicillin with 100 µl of E. coli competent cells.

Uses of Each Strain
- E. coli HB101: Stable genotype; used as a general-purpose strain for recombinant DNA experiments.
- E. coli JM109: Used for alpha-complementation (blue/white screening) and ssDNA preparation; ideal for library construction and subcloning.
- E. coli CJ236: Used for preparation of uracil-containing ssDNA (Kunkel’s method); cultivation requires chloramphenicol for F` episome maintenance.
- E. coli BMH71-18 mutS: Used for site-directed mutagenesis; lacks mismatch repair capability, increasing mutagenesis efficiency.
- E. coli MV1184: Used to select amber mutants; suitable as M13 phage or phagemid vector host for ssDNA preparation.
- E. coli HST02: Restriction-minus phenotype allows efficient cloning of methylated DNA; suitable for alpha-complementation and library preparation.
- E. coli DH5α: General-purpose cloning strain; economical blue/white screening without IPTG.
- E. coli HST08: Lacks restriction genes (mrr, hsd_RMS, mcr_BC, mcrA); recommended for cloning large (>10 kb) or methylated DNA fragments.
- E. coli HST16CR: Similar to HST08; optimized for cloning methylated or large DNA fragments.

Stability of F` Plasmid and α-Complementation
- JM109, BMH71-18 mutS, MV1184, HST02: Less than 1% white colonies when transformed with pUC19 or pUC119 and plated on L-agar with ampicillin, IPTG, and X-Gal.
- CJ236: Strains lacking F` plasmid (pCJ105) do not grow on chloramphenicol plates.
- DH5α: Blue colonies appear when transformed with pUC19 and plated on L-agar with ampicillin and X-Gal.
