Product Specific Information

The SK1 monoclonal antibody reacts with human CD8a, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8α/β) or homodimer (CD8α/α). CD8a is found on thymocytes, mature T cells, and NK cells. It binds MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.

Super Bright 702 is a tandem dye excited at 405 nm and emitting at 702 nm. Recommended filter: 710/50 bandpass. Ensure instrument compatibility.

When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein crucial for T-cell signal transduction and activation. It forms disulfide-linked heterodimers (CD8α/β) acting as co-receptors for MHC class I molecules. CD8α is essential for MHC-I binding and is expressed on T cells, NK cells, monocytes, and dendritic cells.

Binding of MHC-I/peptide complexes triggers Lck recruitment, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are essential for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
