Product Specific Information

The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor and member of the immunoglobulin gene superfamily.
CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, natural killer cells, a subset of normal B cells, and malignant B cells.
Cross-linking of CD7 modulates T cell and NK cell activity through calcium flux, adhesion molecule expression, cytokine secretion, and proliferation.
CD7 associates with phosphoinositol 3'-kinase and induces D-3 phosphoinositide production and tyrosine phosphorylation.

A clonogenic subpopulation of CD34(+) CD38(-) cord blood cells expressing CD45RA, HLA-DR, and high CD7 levels can generate B cells, NK cells, and dendritic cells, distinguishing primitive lymphoid progenitors from pluripotent stem cells.
CD7 also cooperates with CD28 in Treg function; double-deficient mice show reduced Treg numbers and suppressive activity.

Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.125 µg)/test.
Laser: Violet (405 nm); Emission: 702 nm

Super Bright 702 is a tandem dye excited at 405 nm and emits at 702 nm. Use a 710/50 bandpass filter.
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.
Light-sensitive: Protect vial and stained samples from light.
Fixation: Samples can be stored up to 3 days at 4°C in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness and FRET efficiency.

Target Information

CD7 (gp40, Leu9) is a 40 kDa immunoglobulin superfamily member.
Its N-terminal (aa 1–107) is homologous to Ig kappa light chains, while the C-terminal domain is proline-rich, forming a stalk structure.
Expressed on immature and mature T cells, NK cells, and some B cell subsets, CD7 modulates immune activation and is a key marker in leukemia diagnostics.
