Product Specific Information

Description:
The UCHT2 monoclonal antibody reacts with human CD5, a 67 kDa protein expressed by most thymocytes, mature T cells, and a subset of B cells. CD5 signaling activates T cells and interacts with CD72 on B cells, playing a key role in T-B cell interaction and proliferation. The UCHT2 clone recognizes primate CD5.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested for flow cytometric analysis of normal human peripheral blood cells.
Recommended usage: 5 µL (0.125 µg) per test in a final volume of 100 µL. Cell number may vary from 10⁵ to 10⁸ cells/test. Optimize titration for best results.

Fluorochrome Information:
Super Bright 702 is a tandem dye excited with a 405 nm violet laser and emits at 702 nm. Recommended detection with a 710/50 bandpass filter. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vials and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days in the dark at 4°C with minimal loss of brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

CD5 is a 67 kDa single-chain transmembrane glycoprotein expressed on mature T-lymphocytes, most thymocytes, and many T-cell leukemias and lymphomas. It also reacts with a subset of activated B cells and may act as a receptor regulating T-cell proliferation. CD5 is found on approximately 95% of thymocytes and 72% of peripheral blood lymphocytes, with strong reactivity in T-cell areas of lymph nodes. CD5 expression is associated with T-cell leukemias, lymphomas, and activated T cells. Diseases linked to CD5 dysfunction include thymus cancer and Richter’s Syndrome.
