Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8αβ) or homodimer (CD8αα). CD8a is expressed on most thymocytes and subsets of mature T and NK cells. It binds to MHC class I molecules and associates with protein tyrosine kinase p56lck, playing a critical role in T-cell development and activation.

This antibody is optimized for flow cytometric analysis and is pre-diluted for use at 5 µL (0.125 µg) per test (100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test).

Handling Notes:

• Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright dyes to minimize non-specific interactions.
• Protect from light; dye is sensitive to photo-induced oxidation.
• Fixed samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor with T cell receptor (TCR) for MHC class I molecules. CD8 is expressed on cytotoxic T cells, NK cells, monocytes, and dendritic cells, primarily as disulfide-linked heterodimers (CD8αβ) or homodimers (CD8αα). Upon MHC-I/peptide binding, CD8 recruits the tyrosine kinase Lck, leading to T cell activation and cytotoxic response. Activated CD8⁺ T cells play a key role in eliminating pathogens and tumor cells, with differentiation enhanced by IL-2, IL-12, and TGF-β1.
