Product Specific Information

Description:
The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily. CD22 is a type I transmembrane glycoprotein composed of two polypeptide chains (CD22α and CD22β, 130–140 kDa) produced by alternative splicing. It is highly expressed on mature B cells and B cell lymphomas.

The extracellular portion contains seven Ig-like domains capable of binding sialic acid moieties on various cell types. The intracellular portion includes six tyrosine residues within ITIM and activation-like motifs phosphorylated upon B-cell receptor engagement, enabling regulation of B-cell receptor signaling.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Recommended use: 5 µL (0.02 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Recommended viability dyes: Invitrogen LIVE/DEAD Fixable Dead Cell stains.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample before combining with cells.
    • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures for optimized fluorescent signatures minimizing spillover and spread.

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in the cytoplasm and membrane of B-lymphocytes. It appears early in B-cell differentiation, similar to CD19. CD22 expression is limited to mature B cells and plasma cells, aiding in leukemia phenotyping.

Expression Profile:

  • Strongly expressed in hairy cell leukemia
  • Positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma
  • Negative in classical Hodgkin’s lymphoma

CD22 preferentially binds α2,6-linked sialic acid. Ligand-induced tyrosine phosphorylation regulates B-cell antigen receptor signaling, acting in both positive and inhibitory pathways via Src family kinases and phosphatases.
