Product Specific Information

The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily.
CD22 is a type I transmembrane glycoprotein composed of two polypeptide chains (CD22α and CD22β, 130–140 kDa) produced by alternative splicing. It is expressed at high levels on mature B cells and B cell lymphomas.

The extracellular domain contains seven Ig-like domains capable of binding sialic acid-containing ligands, while the intracellular domain includes six tyrosine residues involved in B-cell receptor signaling regulation.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Suitable for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended usage: 5 µL (0.2 µg) per 100 µL test sample (10⁵–10⁸ cells/test).

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
    • Add 5 µL per sample to antibody cocktails prior to cell incubation.
    • For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor Technology:
Based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for optimized fluorescence with minimal spillover and spread.

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in the cytoplasm and membrane of B-lymphocytes.
It appears early in B-cell differentiation, similar to CD19, and is mainly expressed in mature B cells and certain leukemias (e.g., hairy cell leukemia).

  • Useful for phenotyping mature leukemias
  • Strongly expressed in hairy cell leukemia
  • Positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma
  • Negative in classical Hodgkin’s lymphoma
  • Binds preferentially to α2,6-linked sialic acid residues
