Product Specific Information

Description:
The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily. CD22 is a type I transmembrane glycoprotein composed of two polypeptide chains (CD22α, CD22β; 130 and 140 kDa) produced by alternative splicing. It is highly expressed on mature B cells and B cell lymphomas. The extracellular region contains seven Ig-like domains capable of binding sialic acid ligands, while the intracellular region contains ITIM and activation-like motifs that regulate B-cell receptor signaling.

Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported:

  • Reported for use in flow cytometric analysis.

Applications Tested:

  • Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
  • Recommended use: 5 µL (0.2 µg) per test in a final volume of 100 µL.
  • Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample to antibody cocktails before combining with cells.
    • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for optimal spectral properties and minimal spillover.

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in the cytoplasm and membrane of B-lymphocytes. It appears early in B-cell differentiation, similar to CD19, and is expressed in mature B cells but absent in plasma cells. CD22 is useful for phenotyping mature leukemias and is strongly expressed in hairy cell leukemia, diffuse large B-cell lymphoma, and nodular lymphocyte predominance Hodgkin’s lymphoma (negative in classical Hodgkin’s lymphoma).
It binds preferentially to α2,6-linked sialic acid and participates in B-cell receptor signaling through phosphorylation of tyrosine residues, acting in both activation and inhibition pathways.
