This BCL-DWN monoclonal antibody reacts with mouse Bcl-6 (B-cell lymphoma 6), a transcriptional repressor abundantly expressed in germinal center B cells and activated B cells. Bcl-6 is essential for B cell proliferation and survival, and plays a role in pre-B cell survival during development. It is constitutively expressed in DLBCLs, Burkitt’s lymphoma, and non-Hodgkin’s lymphoma, as well as in CD4+ T follicular helper and memory T cells. Bcl-6 expression is antagonized by Blimp-1.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of splenocytes from SRBC-immunized mice. Recommended ≤0.015 µg/test (100 µL volume). Cell number range: 10⁵–10⁸ cells/test. Optimize by titration for best results.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by a 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable). Ensure instrument compatibility.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Store samples up to 3 days in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) at 4°C in dark.
Excitation: 488 nm; Emission: 710 nm; Laser: Blue.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Bcl-6 proto-oncogene encodes a Kruppel-type zinc-finger protein mainly expressed in germinal center B cells and related lymphomas. It is involved in chromosome rearrangements at 3q27 in non-Hodgkin’s lymphomas, detected in 33–45% of diffuse large B cell lymphomas. Bcl-6 expression is observed in follicular, diffuse large B cell, and Burkitt’s lymphomas, as well as in nodular lymphocyte predominant Hodgkin’s disease.
