Product Specific Information

The 8.1.1 monoclonal antibody reacts with mouse podoplanin (T1a, gp38, aggrus), a 43 kDa transmembrane glycoprotein expressed in kidney glomerular epithelial cells (podocytes). Podoplanin is also found in epithelial and mesothelial cells such as intestinal epithelium, alveolar type I cells, and mesothelium of the visceral peritoneum. It serves as a potent marker for lymphatic endothelium and is expressed by subcapsular epithelial cells of the murine thymus. Mice deficient in Podoplanin die at birth due to respiratory defects and congenital lymphedema.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of TE-71 cell line at ≤0.25 µg/test.
A test is defined as the amount of antibody staining a cell sample in 100 µL final volume. Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best performance.

Fluorochrome Information:
PerCP-eFluor 710 emits at 710 nm and is excited by the 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure the instrument can detect this fluorochrome.

Light Sensitivity: This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

Podoplanin (PDPN, T1A, gp38, Aggrus) is a mucin-type glycoprotein (40–43 kDa) expressed in many tumors and normal cells, especially lymphatic epithelial cells and follicular dendritic cells. It localizes in stromal cells of peripheral lymphoid tissue and thymic epithelial cells. Podoplanin regulates lymphatic endothelium and maintains podocyte shape. It acts as a ligand for CLEC-2 and is associated with invasive and metastatic tumor potential. It also contributes to cell migration, metastasis formation, and tumor invasion.

Podoplanin expression is induced via Src activation of Cas and has been observed on Th17 cells and a small subset of CD4+ human PBMCs. Dysfunction is linked to diseases such as lymphangioma and subglottic neoplasm. It is also proposed as a marker for lung injury.
