Product Specific Information

The M5/114.15.2 monoclonal antibody reacts with mouse MHC class II molecules (I-A and I-E subregions). It recognizes glycoproteins such as I-A^b, I-A^d, I-A^q, I-E^d, I-E^k, but not I-A^f, I-A^k, or I-A^s.
This antibody detects polymorphic determinants on B cells, monocytes, macrophages, dendritic cells, and activated T lymphocytes from mice with H-2^b, H-2^d, H-2^q, H-2^p, H-2^r, and H-2^u haplotypes, but not H-2^s or H-2^f.

The M5/114 mAb inhibits I-A-restricted T cell responses in mice with H-2^b, H-2^d, H-2^q, and H-2^u haplotypes.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse spleen cells. Recommended ≤0.06 µg/test (100 µL final volume). Optimize empirically for best performance.

Fluorochrome Information:
PerCP-eFluor® 710 can replace PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5. Emits at 710 nm, excited by 488 nm blue laser.
Recommended filters: 685 LP dichroic mirror with 710/40 bandpass (or 695/40 acceptable).
Stable in samples fixed with 2% formaldehyde overnight at 4°C.
Excitation: 488 nm; Emission: 710 nm; Laser: Blue.
Post-manufacturing filtration: 0.2 µm.

Target Information

MHC class II molecules are transmembrane glycoproteins expressed on antigen-presenting cells (macrophages, dendritic cells, B cells). They bind exogenous antigens in endosomes and present them to T cells.
The peptide-binding groove between MHC class II α and β chains is open at both ends (15–24 amino acids long).
HLA and MHC antibodies are widely used in immunopeptidomics for neoantigen identification via LC-MS/MS.
