Product Specific Information

The monoclonal antibody 2D3 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein involved in adhesion via interactions with SIRP and integrins (CD51/CD61, CD41/CD61). It mediates bi-directional signaling that affects neural synaptic activity and macrophage phagocytosis. CD47 also serves as a receptor for thrombospondin.

Expression is found in most hematopoietic cells (T, B cells, monocytes, platelets, erythrocytes) and non-hematopoietic cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested with normal human peripheral blood cells.
Use at 5 µL (0.6 µg) per test in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) should be used for optimal staining with NovaFluor conjugates. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures optimized for reduced spillover and spread.

Target Information

CD47 (integrin-associated protein, IAP) is a cell surface transmembrane glycoprotein that interacts with integrins and regulates signaling pathways, including G protein activation and calcium flux during adhesion. It binds thrombospondin and participates in membrane transport and signal transduction. CD47 acts as a ligand for CD172a (SIRPα), inhibiting macrophage phagocytosis of CD47-positive cells. It influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, being abundant in synapse-rich regions of brain and retina.
