Product Specific Information
This antibody was cross-adsorbed to a phospho-tyrosine peptide before affinity purification using phospho-p38alpha (Tyr-323) peptide (without carrier).
The antibody detects a 38 kDa band corresponding to p38alpha on SDS-PAGE immunoblots of human Jurkat, K562, and mouse macrophage (J774A.1) cells treated with pervanadate.
Additionally, it detects recombinant p38 MAP Kinase when treated with Fyn kinase (in vitro kinase model provided by Dr. Paul Mittelstadt, Laboratory of Immune Cell Biology, NCI).
Target Information
MAPK14 (p38 alpha) is a member of the stress-activated protein kinase class of MAPKs. The p38 MAPK cascade regulates various cellular responses to stress, inflammation, and other signals.
p38 MAPK is relatively inactive in the nonphosphorylated form and becomes rapidly activated by dual phosphorylation of a Thr-Gly-Tyr motif.
There are four isoforms of p38 MAPK that differ in tissue expression and affinity for upstream activators and downstream effectors.
Upon exposure to tumor necrosis factor, interleukin-1, heat shock, or other activating stimuli, MAPK kinase-3 becomes phosphorylated and activates MAPK kinase-3/6, which in turn phosphorylate Thr180 and Tyr182 residues in p38 MAPK.
Activated phospho-p38 MAPK phosphorylates transcription factors such as ATF-2, CHOP-1, and MEF-2.
