Target Information
TAK1 (MAP3K7)는 세린/트레오닌 키나아제 계열에 속하며 다양한 신호 전달 기능을 수행합니다. TGF-β 활성화 키나아제로 처음 발견되어 TGF-β 수용체 및 TRAF6과 상호작용하여 JNK 및 p38을 조절합니다. 또한 Wnt, BMP, activin, 갑상선 호르몬, 삼투 스트레스, endothelin, ephrine 등의 신호 경로에서도 p38과 JNK의 활성화에 관여합니다.
TAK1은 von Hippel-Lindau 종양 억제 유전자 발현을 통해 PDGF-B, integrin β1, β5를 억제하여 상처 치유를 촉진합니다. 또한 IL-1, IL-6, IL-18, TNF, CD40, TLR, RIG-I 등 다양한 경로에서 IKK 활성화에 필수적인 역할을 합니다. TAK1-IKK 경로 활성화에는 TAB2/3 및 유비퀴틴화가 필요하며, TAK1은 Thr178, Thr184, Thr187, Ser192에서 자가 인산화되고 이후 IKK-β를 인산화합니다.
추가적으로 TAK1은 인간 텔로머레이스 역전사효소를 억제하여 세포 수명 조절에 기여합니다.
