Product Specific Information

The 390 monoclonal antibody reacts with mouse CD31 (PECAM-1, gpIIa), a 130–140 kDa surface protein expressed by endothelial cells and at low levels on all leukocytes and platelets. CD31 plays a key role in adhesion, cell-cell and cell-matrix interactions, and signal transduction.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:

• Flow cytometric analysis
• Immunohistochemical staining of frozen tissue sections

Applications Tested:

• Flow cytometric analysis of mouse thymocytes and splenocytes (≤ 0.6 µg/test)
• Immunohistochemistry of frozen tissue (≤ 20 µg/mL)
• Optimize antibody titration for best performance

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample or per 100 µL of cell suspension (10³–10⁸ cells).
• Add CellBlox Plus Blocking Buffer to antibody cocktails before combining with cells.

Technology:
NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures, engineered for optimized fluorescence with minimized spillover and spectral spread.
Excitation: 509 nm; Emission: 665 nm; Laser: 488 nm (Blue).

Target Information

CD31 (PECAM-1) is an inhibitory coreceptor regulating T and B cell signaling via ITIM motifs. It is expressed throughout the vascular system, mainly at cell junctions. The N-terminal Ig-like domain mediates homophilic binding, critical for cell-cell interactions. CD31 functions in integrin-mediated adhesion, transendothelial migration, angiogenesis, apoptosis, immune regulation, and vascular homeostasis.
