Product Specific Information

The SK1 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as either a heterodimer with CD8 beta chain (CD8 alpha/beta) or a homodimer (CD8 alpha/alpha). Most thymocytes and a subset of mature T cells and NK cells express CD8a. CD8 binds to MHC class I and, through association with protein tyrosine kinase p56lck, plays a role in T-cell development and activation.

Each product contains:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.125 µg) per test in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated conjugates ship with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01). This buffer enhances staining performance and should be used in all NovaFluor experiments.

• Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
• For single-color controls: add 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It plays an integral role in signal transduction, T cell differentiation, and activation. CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor for MHC class I molecules. CD8α is essential for MHC-I binding and is also expressed on NK cells, monocytes, and dendritic cells as CD8α homodimers.

Upon ligation of MHC-I/peptide complexes presented by APCs, CD8 recruits Lck kinase, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs play a crucial role in pathogen and tumor cell clearance. Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
