Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 upregulates genes such as p21waf1, contributing to G1 or G2/M arrest.

Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for p53 activation and stabilization, while phosphorylation at Ser392 is associated with human tumor formation. p53 also plays roles in aging, oxidative stress, and neuronal function. Normally expressed at low levels, p53 is overexpressed in many transformed cell lines, contributing to malignancy. Mutations in the TP53 gene can lead to loss of tumor suppressor activity and are implicated in various human cancers, including those seen in Li-Fraumeni syndrome.
