Target Information
Focal Adhesion Kinase (FAK)는 125 kDa의 비수용체 단백질 티로신 키나아제로, Src의 기질로 작용하며 인테그린 신호전달의 핵심 요소입니다. FAK는 세포 확산, 분화, 이동, 세포사 및 세포주기 G1에서 S기로의 전환 가속화에 중요한 역할을 합니다.
FAK는 중심 촉매 도메인과 초점 부착 부위에 위치시키는 C-말단 꼬리를 가지고 있습니다. 이는 세포가 인테그린 수용체를 통해 세포외기질에 부착하는 부위입니다.
Fibronectin과의 인테그린 결합 시 FAK의 티로신 인산화가 증가하며, Grb2 어댑터 단백질 및 c-Src PTK와의 결합을 유도하고 ERK2 MAP kinase의 활성화를 유발합니다.
v-Src 변형 NIH3T3 세포에서는 FAK와 v-Src, Grb2, Sos의 결합이 fibronectin 부착과 무관하게 발생합니다. Grb2 SH2 도메인은 인산화된 FAK의 Tyr925 부위와 직접 결합하며, 여러 전사 변이체가 존재하지만 세 가지의 완전 길이 형태가 확인되었습니다.
