Description:
The CD28.6 monoclonal antibody reacts with the human CD28 molecule, a 44 kDa homodimer expressed by thymocytes, mature T cells, and plasma cells. CD28 is a ligand for CD80 (B7-1) and CD86 (B7-2), acting as a potent costimulator of T cells. Signaling through CD28 enhances IL-2 and IL-2 receptor expression and increases cytotoxicity of CD3-activated T cells. The CD28.6 antibody has been reported for use in immunoblotting (WB), neutralization studies, and co-stimulation assays related to IL-2 production.

Applications Reported:
Reported for use in flow cytometric analysis and immunoblotting (WB). The recommended titer for WB is approximately 1 µg/mL. CD28.6 has also been reported in in vitro functional assays, exhibiting both blocking and co-stimulatory activities.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Can be used at ≤1 µg per test (defined as the amount of antibody staining a sample in 100 µL). Cell numbers may range from 10⁵ to 10⁸ per test. It is recommended to titrate the antibody for optimal performance.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell-specific surface glycoprotein, part of the immunoglobulin superfamily. It is constitutively expressed on most peripheral blood T lymphocytes and functions as a critical co-stimulatory receptor. CD28 provides the essential second activation signal by binding CD80 and CD86 on antigen-presenting cells. It prevents T cells from entering an anergic state or undergoing premature apoptosis. In murine lymphoid organs and blood, all CD4+ and CD8+ T cells express CD28. Expression is highest on immature thymocytes (CD3-, CD8+, CD4+8+, and CD4-8- subsets). CD28 dysfunction is associated with diseases such as mycosis fungoides and Sézary’s disease.
