Description:
The 1C6 monoclonal antibody reacts with canine CD28, expressed by thymocytes, most T cells, and a subset of NK cells. CD28 binds to CD80 (B7-1) and CD86 (B7-2) and serves as a potent costimulator of T cells, enhancing IL-2 and IL-2 receptor expression and cytotoxicity of CD3-activated T cells.

The 1C6 antibody shows both antagonist activity and proliferation induction. The 1C6 and 5B8 antibodies recognize different epitopes of canine CD28.

Applications Reported:
Reported for use in flow cytometric analysis and functional assays. For proliferation assays, use polypropylene plates. (Fluorochrome-conjugated 5B8 is recommended for flow cytometry.)

Applications Tested:
Tested by flow cytometric analysis of normal canine peripheral blood cells. Recommended use ≤1 µg/test in 100 µL final volume. Cell number can range from 10⁵–10⁸ cells/test. Optimize antibody titration for best results.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell surface glycoprotein of the immunoglobulin superfamily. It is constitutively expressed on most peripheral blood T lymphocytes and acts as a critical T cell costimulatory receptor by binding CD80 and CD86 on antigen-presenting cells. CD28 signaling prevents T cells from entering an anergic state or undergoing premature apoptosis. In mice, CD28 is expressed on all CD4⁺ and CD8⁺ T cells, with expression levels varying by maturation stage. CD28 dysfunction is associated with diseases such as mycosis fungoides and Sezary’s disease.
