The AREG559 monoclonal antibody reacts with human Amphiregulin (AR), a member of the Epidermal Growth Factor (EGF) family. AR is expressed as a transmembrane precursor protein and released by proteolytic cleavage. Soluble AR binds to the EGF receptor (EGFR), promoting growth of normal epithelial cells, fibroblasts, and keratinocytes.
In the immune system, AR acts as an effector molecule for tissue repair and homeostasis, mediating resistance and tolerance to infection and suppressing inflammation. Under various disease or activation conditions, AR is expressed in T cells, group 2 innate lymphoid cells, basophils, mast cells, eosinophils, and neutrophils.

Applications Tested: Tested by western blot analysis of stimulated normal human peripheral blood cells (recommended 1–10 µg/mL). Optimize dilution for best assay performance.
Target Information
Amphiregulin (AR) binds to EGFR with lower affinity than EGF. The mature, secreted AR is an 84-amino acid glycosylated polypeptide growth regulator, derived by proteolytic processing of a 252-amino acid transmembrane precursor.
Seven distinct polypeptide ligands bind to the extracellular domain of EGFR: EGF, TGFα, AR, HB-EGF, cripto-1, epiregulin, and betacellulin. These ligands share a characteristic EGF-like domain defined by six cysteine residues forming three disulfide-linked loops.
AR protein is localized in epithelial tissues of the colon, stomach, pancreas, breast, and placenta, and is reportedly overexpressed in human cancers of breast, colon, stomach, and pancreas.
