Product Specific Information

Description:
The hNanog.2 antibody recognizes human Nanog, a homeobox transcription factor expressed in embryonic stem cells (ESC). Nanog is essential for ESC self-renewal and inner cell mass maintenance, functioning similarly to Oct4 and Sox2. It regulates proliferation and prevents differentiation by directly binding DNA consensus sequences to activate or repress transcription. In differentiating ESCs, Nanog binds to its own promoter to repress expression. Overexpression of Nanog has been reported in germ cell tumors and certain cancers such as breast and colorectal cancer.

Applications Reported:
This antibody has been reported for use in:

  • Western blotting
  • Immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections
  • Immunocytochemistry

Applications Tested:

  • Western Blot: ≤ 1 µg/mL using NTERA cell lysates
  • Immunohistochemistry (Paraffin): ≤ 20 µg/mL with high or low pH antigen retrieval
  • Immunocytochemistry: ≤ 5 µg/mL on formaldehyde-fixed and permeabilized cells

It is recommended to titrate the antibody for optimal performance in each assay.

Target Information

NANOG (Nanog homeobox):
NANOG is a homeodomain protein that regulates pluripotency and differentiation in embryonic stem cells. NANOG mRNA is expressed in pluripotent human and mouse cell lines but absent in differentiated cells. Human NANOG shares 52% amino acid identity with mouse NANOG (85% in the homeodomain).

High NANOG expression is observed in undifferentiated N-Tera embryonal carcinoma cells. NANOG acts as a transcription regulator promoting self-renewal and proliferation of embryonic stem cells. Its role in embryonic development suggests potential in regenerative medicine and induced pluripotent stem (iPS) cell generation, where NANOG, along with Sox2, Klf4, and c-Myc, can induce pluripotency without viral vectors. Overexpression of NANOG promotes S-phase entry and cell proliferation.
Diseases associated with NANOG dysfunction include teratocarcinoma and germ cell/embryonal cancers.
